Hi there,
First, make your way to Shin-Osaka station, this can be by local train, metro, taxi or your choice of transport. Note, that the JR Pass will only cover local JR lines.
From Shin-Osaka, take a Hikari Shinkansen to Tokyo, and there transfer to the Hayabusa Shinkansen to Shin-Hakodate-Hokuto, the Shinkansen station for Hokkaido. Finally, take a fast tld express train to Sapporo.
