Yes, the JR pass is valid for this route on the Sunrise Seto or Izumo sleeper trains - as a rail pass holder you can use the nobi nobi carpeted areas free of charge. As these areas can fill up quickly, make sure you reserve your places on this train as soon as you land at the JR ticket office in Narita.

As an alternative, If you can't get on the night train for whatever reason, you still have the alternative of booking and using the [JR Kanto night bus to Osaka] with your Japan Rail Pass, so you can book this at Narita and board at Tokyo Station.

Do u think it is advisable to purchase the rail pass. As we are only travelling within this 2 places and if we are getting the rail pass. We hope to maximize the usage within Osaka and Tokyo.

Tokyo>Osaka on the Sunrise Seto or Izumo Sleeper is ¥14,870. Combine that with your return to Tokyo by bullet of ¥13,950 and at least one of your Narita Express legs of ¥3140, and you have a total minimum spend of ¥31,960.

This makes it well worth your while to purchase a 7 day rail pass for this journey. You will also have flexibility in Osaka and Tokyo to do day trips should you want to, and will have free use of the JR lines within Osaka and Tokyo.
