If I hold the JR East–South Hokkaido Rail Pass, may I reserve a Gran Class seat on the Shinkansen from Tokyo to Shin-Hakodate-Hokuto?
If it is possible, could you please inform me of the additional cost required for this route?

Hi there,
You can't reserve Gran or Green Class seats using the JR East–South Hokkaido Rail Pass.
You'd have to pay the full Shinkansen and Gran/Green Class fare, but that that point the value of the JR East–South Hokkaido Rail Pass would be so bad, that it would be better to just buy the tickets normally.
